Biological Activity

PF-05221304 is an orally bioavailable, liver-targeted inhibitor of acetyl-CoA carboxylase (ACC), which plays a key role in lipid metabolism. By inhibiting ACC, the compound suppresses the initial committed step of hepatic de novo lipogenesis (DNL), thereby decreasing fatty acid synthesis and suppressing hepatic lipid accumulation. This targeted modulation of lipogenic activity attenuates triglyceride accumulation and cellular steatosis in liver tissue. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
