Biological Activity

PF-04929113 (SNX-5422) is an orally bioavailable prodrug of SNX-2112, a potent and selective heat shock protein 90 (HSP90) inhibitor. Upon conversion to SNX-2112, it binds HSP90 and disrupts its chaperone activity, promoting the proteasomal degradation of oncogenic client proteins such as HER2 and AKT. This target inhibition downregulates downstream cell survival pathways, ultimately inducing cell cycle arrest, apoptosis, and suppression of tumor cell proliferation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
