PF-04457845 is a potent, selective fatty acid amide hydrolase (FAAH) inhibitor targeting human FAAH (IC50 7.2 nM) and rat FAAH (IC50 7.4 nM). It covalently modifies the active-site serine nucleophile of FAAH with exquisite selectivity over other serine hydrolases. By blocking FAAH-mediated degradation of endocannabinoids, PF-04457845 elevates anandamide levels to suppress nociceptive and inflammatory signaling pathways. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
