PD 150606 is a selective, cell-permeable calpain inhibitor of µ-calpain (Ki = 0.21 µM) and m-calpain (Ki = 0.37 µM). By blocking calcium-dependent cysteine protease activity, it suppresses pathological proteolysis and downstream apoptotic cell death pathways. In neuronal disease models, PD 150606 effectively reduces hypoxic and hypoglycemic injury in cerebrocortical neurons while protecting cerebellar Purkinje cells against excitotoxic damage. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
