Biological Activity

Zelenirstat (PCLX-001) is an orally active dual N-myristoyltransferase (NMT) inhibitor targeting NMT1 and NMT2. Treatment with PCLX-001 impairs global protein N-myristoylation, suppressing early B-cell receptor (BCR) signaling pathways essential for cell survival and inducing apoptosis in lymphoma models. Furthermore, inhibition of NMT disrupts mitochondrial respiratory complex I assembly by downregulating NDUFAF4, leading to the loss of oxidative phosphorylation and cellular respiration. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
