PCI-34051 is a potent and selective histone deacetylase 8 (HDAC8) inhibitor with an IC50 of. It exhibits over 200-fold selectivity for HDAC8 over HDAC1 and HDAC6, and more than 1000-fold selectivity over HDAC2, HDAC3, and HDAC10. PCI-34051 induces mitochondrial cytochrome c release and caspase-dependent apoptosis, demonstrating selective cytotoxic activity against T-cell malignancy cell lines without causing widespread histone or tubulin hyperacetylation.
