PD-0332991 (Palbociclib) Isethionate is a highly selective inhibitor of CDK4 (IC50 = and CDK6 (IC50 = in cell-free assays, showing no inhibition against CDK1, CDK2, CDK5, or receptor tyrosine kinases. By blocking the cyclin D-CDK4/6 complex, it prevents retinoblastoma protein phosphorylation, downregulating E2F-mediated transcription. This pathway inhibition effectively blocks G1 to S phase cell cycle transition, resulting in robust cell cycle arrest in Rb-positive tumor cells. [1]
