Palbociclib 2HCl is a highly specific inhibitor of cyclin-dependent kinase 4 (CDK4, and CDK6. By blocking the CDK4/6-cyclin D complex, it suppresses retinoblastoma (Rb) protein phosphorylation at Ser780 and Ser795, preventing downstream cell cycle progression. This selective inhibition effectively blocks signal transduction, inducing G1 phase cell cycle arrest and suppressing proliferation in Rb-positive tumor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
