P22077 is a small-molecule inhibitor of ubiquitin-specific protease USP7 that also inhibits the closely related deubiquitinase USP47. By blocking USP7 activity, P22077 stabilizes p53 levels, reduces MDM2 expression, and suppresses tumor growth and survival signaling across various cancer models. Furthermore, USP7 inhibition impairs DNA damage repair pathways and induces cell cycle arrest and apoptotic cell death in treated cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
