Biological Activity
ON123300 is a potent, ATP-competitive multi-targeted kinase inhibitor targeting CDK4, Ark5 (NUAK1), RET, Fyn, PDGFRβ, and FGFR1. By co-targeting CDK4 and Ark5, ON123300 effectively disrupts cell cycle progression through RB phosphorylation blockade and inhibits downstream AKT/mTOR signaling pathways. Consequently, this compound suppresses tumor cell proliferation, impairs cellular survival, and induces apoptotic cell death across preclinical cancer models, including glioblastoma. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).

Pre-aliquoted for Longevity: Solid-state aliquots offer longer shelf life compared to stock solutions.
