Biological Activity

Olutasidenib is a potent, selective, and brain-penetrant inhibitor of mutant IDH1-R132H and IDH1-R132C. By blocking mutant IDH1 activity, it reduces the production of the oncometabolite D-2-hydroxyglutarate (2-HG) in mutant IDH1-expressing tumor cells. Lowering 2-HG levels reverses histone and DNA hypermethylation, which restores normal cell differentiation pathways and inhibits proliferation in mutant IDH1-driven cancers. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
