Biological Activity

Olomorasib (LY3537982) is a potent and highly selective second-generation small-molecule inhibitor of GDP-bound KRAS G12C. By preferentially binding to the inactive, GDP-bound state of mutant KRAS G12C, it effectively blocks interaction with downstream effectors and impairs the RAF/MEK/ERK signaling pathway. This target inhibition leads to downstream cell cycle arrest and suppression of cellular proliferation in mutant KRAS G12C-bearing tumor cells. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
