Biological Activity

Oclacitinib maleate is an inhibitor of the Janus kinase (JAK) family, targeting JAK members with IC50 values ranging from 10 to 99 nM. It selectively suppresses JAK1-dependent cytokine signaling with IC50 values between 36 and 249 nM, without inhibiting a panel of 38 non-JAK kinases. By blocking JAK family activity, it inhibits downstream STAT protein phosphorylation, thereby suppressing pro-inflammatory cytokine-mediated signaling pathways and cellular immune responses.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
