Biological Activity
NX-2127 is an orally bioavailable targeted protein degrader of Bruton's tyrosine kinase (BTK). By recruiting the cereblon E3 ubiquitin ligase complex, NX-2127 induces the ubiquitylation and proteasomal degradation of BTK, suppressing downstream B-cell receptor (BCR) signaling pathways. Concomitantly, it mediates degradation of the transcription factors IKZF1 and IKZF3, thereby stimulating T-cell activation and enhancing IL-2 production in primary human T cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
