NVP-LCQ195 is a small molecule heterocyclic inhibitor of cyclin-dependent kinases, including CDK1, CDK2, CDK3, CDK5, and CDK9. By inhibiting CDK activity, NVP-LCQ195 induces cell cycle arrest and apoptotic cell death in multiple myeloma cells at sub-micromolar concentrations while sparing non-malignant cells. Downstream, it suppresses transcriptional signatures associated with oncogenesis, drug resistance, and stem cell renewal, including MYC, HIF-1α, and IRFactivation, and overcomes bone marrow microenvironmental protection. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
