Biological Activity

NT-0527 is a potent, brain-penetrant inhibitor of the NLRP3 inflammasome. By selectively targeting NLRP3, it inhibits inflammasome assembly and downstream activation of inflammatory cascades. This inhibition blocks the release of pro-inflammatory cytokines such as IL-1β and IL-18, mitigating neuroinflammation and systemic inflammatory responses in animal models. NT-0527 exhibits oral bioavailability and demonstrates potent anti-inflammatory efficacy in vivo.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
