NSC16168 is a specific inhibitor of ERCC1-XPF endonuclease activity, exhibiting an IC50 value of 0.42 μM in a high-throughput screening assay. By suppressing ERCC1-XPF function, the compound impairs nucleotide excision repair (NER) pathways required for clearing platinum-induced DNA lesions. This inhibition blocks DNA repair mechanisms, thereby sensitizing cancer cells and potentiating cisplatin cytotoxicity and antitumor efficacy. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
