NSC 185058 is a selective small-molecule inhibitor of the cysteine protease ATG4B. By blocking ATG4B activity, it prevents the lipidation of LC3B and suppresses cellular autophagy without perturbing the mTOR or PtdIns3K signaling pathways. Disruption of autophagic flux by NSC 185058 leads to reduced cell viability and impairs tumor growth in preclinical models of aggressive breast cancer.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
