NS8593 is a potent negative gating inhibitor of small-conductance Ca2+-activated K+ channels, including SK1, SK2, and SK3 (KCa2.1–2.3). By reversibly shifting the gating calcium sensitivity of SK channels, NS8593 suppresses potassium currents and modulates membrane potential without blocking the ion conduction pore. This targeted inhibition disrupts calcium-dependent hyperpolarization pathways, altering neuronal excitability and cellular electrophysiology. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
