Biological Activity

NS3623 is a small-molecule activator of human ether-a-go-go-related gene (hERG1/Kv potassium channels that exhibits a dual mode of action by both activating and inhibiting hERG1 channels. Activation of hERG1 enhances rapid delayed rectifier (IKr) and transient outward (Ito) potassium currents, driving cellular repolarization. Downstream, NS3623 shortens cardiac action potential duration, improves repolarization reserve, and displays antiarrhythmic phenotypic effects. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
