NS309 is a potent activator of small-conductance and intermediate-conductance calcium-activated potassium channels, while exhibiting no activity against large-conductance potassium channels. By enhancing these calcium-activated potassium currents, NS309 shifts channel gating to lower calcium concentrations and induces membrane hyperpolarization. This hyperpolarization subsequently suppresses voltage-gated calcium channel activity, resulting in reduced cellular excitability and attenuated smooth muscle contraction. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
