NPD4928 is a small-molecule inhibitor of ferroptosis suppressor protein 1 (FSP1) that acts as a potent ferroptosis enhancer. By blocking FSP1 enzymatic function, NPD4928 disrupts glutathione-independent antioxidant protection and significantly sensitizes diverse cancer cell lines to glutathione peroxidase 4 (GPX4) antagonists, thereby driving lipid peroxidation and promoting synergistic cytotoxic cell death through the ferroptosis signaling pathway. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
