Novobiocin is an orally active aminocoumarin antibiotic that acts as an inhibitor of bacterial DNA gyrase and eukaryotic topoisomerase II, as well as an antagonist of heat shock protein 90 (Hsp90). By targeting the ATP-binding site of DNA gyrase B and the C-terminal domain of Hsp90, it disrupts bacterial DNA supercoiling and induces degradation of Hsp90 client proteins, leading to cell cycle arrest and antimicrobial activity. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
