NMDIis a small-molecule inhibitor of nonsense-mediated RNA decay (NMD) that targets SMG7 to disrupt SMG7–UPF1 interaction. By blocking the interaction between SMG7 and UPF1 within the NMD pathway, NMDIincreases the stability and expression of NMD-targeted mRNAs harboring premature termination codons. Consequently, this inhibition restores full-length protein production in cells containing nonsense-mutated p53, thereby promoting tumor suppressor activity. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
