Biological Activity

NiCur is a small-molecule inhibitor of CREB-binding protein (CBP) histone acetyltransferase (HAT) activity. By blocking CBP HAT function, NiCur downregulates p53 activation following genotoxic stress. On the CDKN1A promoter, it diminishes the recruitment of p53 and RNA polymerase II while reducing histone H3 acetylation levels. Consequently, NiCur modulates chromatin structure and attenuates p53-directed apoptosis in normal gastrointestinal epithelial cells. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
