Biological Activity
Niacin hydrochloride is a direct and noncompetitive inhibitor of hepatocyte diacylglycerol acyltransferase-2 (DGAT2). By blocking DGAT2, a key enzyme involved in triglyceride synthesis, it accelerates intracellular hepatic apolipoprotein B degradation. This pathway inhibition leads to decreased hepatic assembly and secretion of very low-density lipoprotein (VLDL) and low-density lipoprotein (LDL) particles, thereby suppressing atherogenic lipid accumulation.
