Biological Activity
Nesuparib (JPI-547) is a dual inhibitor of poly(ADP-ribose) polymerase (PARP1/2) and tankyrase (TNKS1/2). By co-inhibiting PARP and tankyrase, Nesuparib suppresses RAD51 expression and impairs homologous recombination repair activity, retarding tumor growth in BRCA-mutated models. Furthermore, it disrupts Wnt/β-catenin and YAP signaling pathways, induces G2/M phase cell cycle arrest, and promotes apoptosis in homologous recombination-deficient tumors. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
