MX69 acts as a small-molecule MDM2/XIAP inhibitor that targets the MDM2 RING domain. By selectively binding to the RING domain of MDM2, MX69 inhibits MDM2 E3 ubiquitin ligase activity and prevents MDM2-mediated protein degradation. Disruption of this signaling axis restores downstream p53 tumor suppressor pathway activity, leading to inhibition of cellular proliferation, cell cycle arrest, and induction of apoptosis in various cancer cell models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
