Biological Activity

MV1035 is an imidazobenzoxazin-5-thione inhibitor targeting the RNA demethylase ALKBH5 and the DNA repair protein ALKBH2. Inhibition of ALKBH5 by MV1035 increases N6-methyladenosine levels and downregulates downstream CD73 protein expression, thereby significantly suppressing glioblastoma U87-MG cell migration and invasiveness. Furthermore, MV1035 inhibits ALKBH2 activity and reduces MGMT expression, acting synergistically with temozolomide to decrease cell viability and sphere formation in glioma stem cells. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
