MTK458 is a synthetic, orally bioavailable, and brain-penetrant small-molecule activator of PTEN-induced kinase 1 (PINK1). Direct binding of MTK458 to PINK1 stabilizes an active kinase complex with the translocase of outer mitochondrial membrane machinery. This activation enhances PINK1 kinase activity, driving downstream mitophagy to facilitate mitochondrial quality control and preserve cellular homeostasis. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
