Biological Activity

MRTX1719 (Navlimetostat) is a potent, selective, MTA-cooperative inhibitor of the PRMT5•MTA complex. By binding selectively to the PRMT5•MTA complex present in MTAP-deleted cancer cells, MRTX1719 suppresses symmetric dimethylarginine (SDMA) protein modification, exhibiting over 70-fold selective inhibition of PRMT5 activity and cell viability in MTAP-deficient cells relative to MTAP wild-type cells. This selective PRMT5 inhibition induces synthetic lethality, driving cell cycle arrest and suppressing tumor growth in MTAP-null cancers. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
