Biological Activity

MPEP is a selective metabotropic glutamate receptor 5 (mGluR5) antagonist with an IC50 of 36 nM. It displays high selectivity for mGluR5 without significant activity at other group I, II, or III metabotropic glutamate receptors. By blocking mGluR5 signaling, MPEP inhibits intracellular calcium mobilization, downstream glutamatergic neurotransmission, and produces robust anxiolytic-like and antidepressant-like phenotypes in rodent behavioral models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
