MLT-748 is a potent and selective allosteric inhibitor of mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1). The compound binds at the structural interface between the caspase and immunoglobulin-like Ig3 domains of MALT1, locking its catalytic domain in an inactive conformation. Inhibition of MALT1 protease activity disrupts downstream NF-kB signaling cascades, thereby attenuating immune cell activation and lymphocyte proliferation.
