MLi-2 is an orally active and highly selective inhibitor of leucine-rich repeat kinase 2 (LRRK2), exhibiting potency in a purified LRRK2 kinase assay (IC50 = 0.76 nM), a cellular assay monitoring LRRK2 pSer935 dephosphorylation (IC50 = 1.4 nM), and a radioligand competition binding assay (IC50 = 3.4 nM). Inhibition of LRRK2 disrupts downstream signaling pathways implicated in Parkinson's disease pathogenesis, providing a potent chemical probe to investigate LRRK2 biology and cellular function. [1]

- Preheat media/saline to 37°C.
