Biological Activity

ML349 is a potent and selective inhibitor of acyl protein thioesterase 2 (APT2) with a Ki of 120 nM and an IC50 of 144 nM. By selectively blocking APT2-mediated depalmitoylation, ML349 disrupts dynamic S-acylation cycles and downstream target trafficking. Consequently, APT2 inhibition by ML349 impairs oncogenic signal transduction, leading to reduced cell proliferation in MCF10A and Snail-overexpressing cell lines.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
