ML335 is a potent and selective activator of two-pore domain potassium channels, specifically TREK-1 (K2P2.1) and TREK-2 (K2P10.1). By directly binding to a regulatory pocket near the selectivity filter, ML335 induces conformational changes in transmembrane helix 6, driving channel activation and enhancing potassium ion conductance. This activation modulates membrane potential and attenuates cellular excitability in potassium channel-regulated signalling pathways. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
