Biological Activity

ML323 is a potent and selective inhibitor of the USP1/UAF1 deubiquitinase complex with an IC50 of. Inhibition of USP1/UAF1 by ML323 promotes the monoubiquitination of key DNA repair proteins, including FANCD2 and PCNA, thereby disrupting the Fanconi anemia pathway and translesion synthesis. Consequently, ML323 impairs DNA damage response mechanisms and sensitizes cancer cells to DNA-damaging chemotherapeutics such as cisplatin. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
