Biological Activity

ML218 is a potent and orally active T-type Ca²⁺ channel inhibitor that targets Cav3.2 (IC50 310 nM) and Cav3.3 (IC50 270 nM), as well as Cav3.1. By blocking voltage-gated calcium influx, ML218 modulates neuronal excitability and suppresses burst activity in subthalamic nucleus neurons. These cellular effects disrupt abnormal firing patterns within central neural circuits, highlighting its function in attenuating hyper-excitable neuronal pathways. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
