Biological Activity

ML216 is a potent and selective inhibitor of Bloom’s syndrome protein (BLM) helicase, displaying DNA unwinding inhibition against BLM 636–1298 (IC50 = 0.97 μM) and full-length BLM (IC50 = 2.98 μM). By disrupting BLM-mediated DNA unwinding activity, ML216 impairs homologous recombination repair and genomic stability pathways. This suppression induces DNA double-strand breaks and suppresses cell proliferation, demonstrating antiproliferative and antitumor activity in cancer models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
