Biological Activity

ML204 is a potent and selective transient receptor potential canonical 4 (TRPC4) channel inhibitor, exhibiting IC50 values of about 1 μM in fluorescent intracellular Ca2+ assays and about 3 μM in whole-cell voltage clamp experiments. By blocking TRPC4-mediated calcium influx, ML204 disrupts downstream calcium-dependent signaling pathways and inhibits TRPC4 channel currents, exhibiting selectivity within the TRPC subfamily and over other non-TRP channels. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
