MKT-077 is a water-soluble rhodacyanine dye that acts as an inhibitor of heat shock cognate 70 (Hsc70) and heat shock protein 70 (Hsp70) family chaperones. By selectively accumulating in the mitochondria of carcinoma cells, MKT-077 disrupts mitochondrial membrane potential and chaperone-mediated protein folding, leading to cell cycle arrest, inhibition of tumor cell proliferation, and induction of apoptosis across various cancer models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
