Biological Activity

MKC8866 is a potent and selective IRE1α RNase inhibitor that suppresses the unfolded protein response (UPR) signaling pathway. By specifically blocking IRE1α endoribonuclease activity, MKC8866 prevents the downstream splicing of XBP1 mRNA, attenuating transcriptional activation of endoplasmic reticulum stress-response target genes. Consequently, inhibition of this pathway by MKC8866 disrupts tumor microenvironment crosstalk, decreases pro-survival signaling, and impairs cancer cell proliferation and survival in preclinical models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
