Biological Activity
MKC-3946 is an inhibitor of the IRE1α endoribonuclease domain. By blocking IRE1α-mediated splicing of XBP1 mRNA, it disrupts the unfolded protein response pathway under endoplasmic reticulum stress. This inhibition leads to attenuated downstream transcriptional programs, triggering growth inhibition and apoptosis in multiple myeloma cell lines while sparing normal mononuclear cells. MKC-3946 also enhances the anti-myeloma activity of proteasome inhibitors such as bortezomib. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
