MK2 Inhibitor III is a potent, ATP-competitive inhibitor of mitogen-activated protein kinase-activated protein kinase 2 (MK2/MAPKAP-K2). By inhibiting MK2 kinase activity downstream of the p38 MAPK signaling pathway, it blocks the phosphorylation of downstream substrates such as heat shock protein 27 (HSP27) and tristetraprolin (TTP). Consequently, this targeted inhibition disrupts the post-transcriptional regulation of pro-inflammatory cytokines, leading to a significant reduction in tumor necrosis factor-alpha (TNF-alpha) expression and cellular inflammatory responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
