Biological Activity

MK-8617 is an orally active pan-inhibitor targeting hypoxia-inducible factor prolyl hydroxylases (HIF PHD1, PHD2, and PHD3). Inhibition of HIF PHD enzymes prevents the oxygen-dependent degradation of HIF-alpha subunits, allowing them to accumulate and translocate to the nucleus. This accumulation activates downstream hypoxic gene expression and promotes erythropoiesis, stimulating red blood cell production and cellular adaptive responses to hypoxia.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
