MK-5108 (VX-689) is an ATP-competitive, selective inhibitor of Aurora A kinase, displaying 220-fold selectivity over Aurora B and 190-fold selectivity over Aurora C. Target inhibition leads to disruption of cell cycle progression, resulting in G2/M phase cell cycle arrest, increased Caspase 3/7 activity, autophagy induction, and suppression of tumor cell proliferation across multiple cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
