Biological Activity

Mirogabalin (DS-5565) is a novel, selective α2δ subunit ligand that targets the α2δ-1 and α2δ-2 subunits of voltage-gated calcium channels. By binding preferentially to the α2δ-1 subunit, it inhibits membrane trafficking of calcium channels and attenuates calcium influx into presynaptic terminals. This reduction in calcium currents suppresses the release of excitatory neurotransmitters in central pain pathways, demonstrating potent and sustained analgesic activity in rodent models of neuropathic pain. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
