Mirabegron is a potent and selective β3-adrenoceptor agonist with an EC50 of 22.4 nM for human β3-adrenoceptors. Activation of β3-adrenoceptors stimulates intracellular cAMP accumulation and downstream protein kinase A signaling. This signaling cascade promotes detrusor smooth muscle relaxation during the urine storage phase, thereby enhancing bladder compliance and increasing voided volume per micturition without reducing contraction force or micturition pressure.
