MIPS521 is a positive allosteric modulator targeting the adenosine A1 receptor (A1R). By binding an extrahelical allosteric site, MIPS521 stabilizes the adenosine-A1R-Gi protein complex, thereby Bio-enhancing G protein-coupled signaling pathways activated by endogenous adenosine. This positive allosteric modulation attenuates nociceptive signaling, producing pronounced in vivo analgesic efficacy in rat neuropathic pain models without significant adverse side effects. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
