SAR405838 is an orally available MDM2 antagonist (Ki 0.88 nM) that selectively inhibits the MDM2-p53 interaction. By disrupting MDM2 binding, SAR405838 blocks p53 degradation and restores p53 transcriptional activity. This leads to the upregulation of downstream target genes such as p21 and PUMA, driving robust cell cycle arrest and apoptosis in p53 wild-type tumor cells.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
